NB: This can be solved by using the normal layering style and using duplicates of the movie footage, rather than using adjustment layers. MJB Dec 2007

This final sample is harder to achieve becuase of the roto'ed subtraction mask. Nevertheless the result is more realistic. The same effect can be actieved by determining the greyscale range which manages the iris and setting this first, then applying others around this problem. NB: Colour hue will need to be taken into consideration as other colours outside of the specificed range will 'wash' over your intended colour (e.g. for the iris) so hueing is necessary for the target area (e.g. for green eyes the orange on the skin may cause hazel eyes, so compensate for this effect with a stronger blue and turn the hue to the obverse of the orange palour for the skin so substract the change to the iris area.)
